I'm going to have to say Lucky☆Star as well. It's one of the few anime titles that truly get me rolling with laughter.
I love a lot of other anime with humor, but not to many of them have humor as the main theme.

Gintama wins hands down. Ouran is more shoujo appeal type of comedy which you can almost find in any ( or almost) every type of drama/comedy and Gintama for shonen types from the action, the dirty jokes and parodies which mostly can only be understand by otakus or about japanese culture.
